The Barbarian: Masters of Battle specialization quest becomes available once your Barbarian hits Level 15. You will need to head to Ked Bardu in the Dry Steppes and speak to Forgemaster Kerdi to begin the quest.
Completing the Masters of Battle quest unlocks the Weapon Technique slot system, which allows you to choose a Weapon Expertise Effect that will be applied to all of your Barbarian's attacks, regardless if that weapon is equipped or not.

The Rogue: True Potential specialization quest will become available once your Rogue hits Level 15. This quest is auto-accepted, and will be set in Fractured Peaks. To begin the quest, speak to an NPC named Leyrana in the town of Menestad. Complete the quest to unlock the Rogue's special class mechanic called the Specialization System.

The Sorcerer: Legacy of the Magi specialization quest will become available once your Sorcerer hits Level 15. This quest is auto-accepted, and will be set in Fractured Peaks. To begin the quest, speak to an NPC named Mordarin found to the southeast of the town of Nevesk.
Complete the Legacy of the Magi quest to unlock the Enchantment Slot system which allows you to equip unlocked spells into Enchantment Slots. Slotted spells will grant powerful modifiers for your build, giving you a power boost after hitting Level 15.

The Druid: Spirits of the Lost Grove sepcialization quest will become available as soons as your Druid hits Level 15. This particular quest can be started by reaching Scosglen. Complete it to unlock the ability to utilize Spirit Animals.

The Necromancer: Call of the Underworld specialization quest will unlock once your Necromancer reaches Level 25. Completing it will expand your Book of the Dead, allowing you to now summon Golems on top of your Skeleton army.
To begin this specialization quest, speak to a ghostly NPC named Maltorius found to the northeast of Menestad. Follow his instructions to fully realize the potential of your Necromancer class.
Necromancer: Call of the Underworld Quest Walkthrough and Rewards
Unlike previous Diablo games, Diablo 4 sets out to make classes feel more unique via the brand new specialization system. Each class in the game will now have their own Class Mechanic that plays to the strengths of their kits, adding new abilities and new effects without having to itemize accordingly (like in previous games).
