The Phase Spider Matriarch can be found in the Whispering Depths, which is located inside the well of the Blighted Village.
To access the Whispering Depths, interact with the well and choose to go down to the depths.

Once inside the Whispering Depths, head east from the well entrance, battling any Phase Spiders in the way, until you find pillars with several spiderweb bridges.
This will signify that the Phase Spider Matriarch is near. You can use Hide to check for the search area of any other enemes in the area.

Using skills or spells that push foes like the Thunderwave, the Phase Spider Matriarch can pushed into the bottomless pits at the sides of the arena.
Keep in mind that you will not be able to loot her if you do this. This is the quickest way to eliminate her, and gives you the Pest Control achievement if you stealth your way through the Phase Spiders and manage to kill her before she spawns her Spiderlings.
One the biggest challenges in facing the Phase Spider Matriarch is the Spiderlings. They can be dangerous if not taken care of quickly, since they can surround and overwhelm one of your party members.
Spells like Thunderwave or Scroll of Burning Hands can be useful in taking out a group of Phase Spider Eggs or Spiderlings. You can also make use of a combo between Grease and fire to quickly decimate them before they get to you.
You can get ahead of the Matriarch spawning the Spiderlings by destroying the Phase Spider Eggs with the above attacks or similar.

The above moves are some examples of moves and bonus moves that you can use to destroy the spiderwebs.
When the Phase Spider Matriarch is on one of the spiderweb bridges, you can destroy that bridge with either a spell or a physical attack to cause the Matriarch to fall and receive a large amount of fall damage.
You can use this to your advantage even when there are Spiderlings present. By also bringing the Matriarch to solid ground, you do not have to worry about being Enwebbed.
When having done enough damage to bring the Phase Spider Matriarch to low HP, it will trigger its Spider Queen's Wrath.
The Spider Squeen's Wrath is a condition that increases the boss's Strength by 2 and reduces its Armor Class by 1. She can can also attack twice in one turn.
This condition will not disappear until she is dead, so make sure to defeat her quickly!

When not yet in battle, crossing the webbed bridge can cause the Enwebbed condition. This will hinder an affected character's movement, and will cause the Phase Spider Matriarch to immediately head towards that character who failed the Saving Throw (akin to real life spiders heading towards trapped prey.)
For this reason, approaching her on the spiderweb bridge at the upper area is not recommended. Instead, if you're at the upper area, use Hide and go down the knotted roots and navigate around lower the area.
Once at the area below, circle around the area (do not head directly to the nearest Phase Spider) and then go around the pit in the middle.
There will be one Phase Spider that is roaming across once you get near the spot with the Dark Amethyst. It will slightly move forward to cover that spot, so wait for it to turn or head to a corner where it cannot cover with its line of sight.
As soon as the Phase Spider turns around, move quickly to the area next to it. There will be cragged rock that you can use to climb and get behind the Phase Spider Matriarch.
Before climbing the cragged rock up to the Phase Spider Matriarch's path, make sure to carefully time when and where it moves to.
Make to sure to attack it behind to initiate a Surprise turn. For a combo opening that does not involve an instant kill, you can attempt to push her onto the spiderweb bridge and have another member destroy it to cause her a great deal of fall damage.
When the Phase Spider Matriarch has been defeated, its body can be looted for the Poisoner's Robe, an uncommon clothing that gives additional 1-4 posion damage to any poison attacks when equipped.
If you wish to loot the Phase Spider Matriarch, make sure you do not kill it by pushing it into the bottomless pit.
